Hello everyone, I am Ethan, the person in charge of Opentrons technical support. This video is the fourth in a series of videos from unboxing to protocol operation. In this episode, let's start connecting the pipette. I just installed the app and connected it to the OT-2 so I could start with the first task, which is connecting the pipette. To do this, I needed the pipette, a 2.5mm hex screwdriver, the pipette screw that comes with the OT-2 or the pipette, and the calibrated aluminum block that comes in the accessory box that attaches to the OT-2 platform, which the OT-2 has 2 holders for Opentrons pipettes. As long as you use Opentrons pipettes, you can connect them in any configuration you want because the OT-2 is very flexible and modular. You'll be amazed how easy it is to connect your pipette to the OT-2! I will show you the pipette connections today using the P1000 and P20 models as a demonstration. Let's start with a computer with our application installed and connected to the OT-2.
First, I'm going to go to the device page and click on the three dots here, click into pipette installation, I'm going to click on the connect pipette button and move the pipette to the front, which will start the connected pipette pipette and bring the pipette holder to the front of the deck with the pipette holder down towards the deck so I can use the pipette holder to attach the pipette. Next I'm going to select the pipette model that I want to connect to the OT-2, I feel like the P1000 is probably the best choice here, I'm going to click on that and select the P1000 single channel pipette.
First I need to align the P1000 pipette with the stand through the front window, I will hold it vertically and the front edge of the pipette should be flush with the front edge of the mounting base. There is nothing here to support the pipette, so if I let go it will fall off. There are 3 and 2.5 mm hex screws that secure the pipette to its holder, there are 2 top screws that are permanently embedded into the clear glass at the top of the base, and I then needed to insert one screw myself. First I'll screw on the top 2 screws, making sure not to tighten them all the way, then I'll unscrew the bottom screws, making sure the pipette is vertical. This time we are installing a single channel pipette so I will unscrew the bottom screw and once the pipette is screwed in I need to plug it into the OT-2 and control the pipette via the white ribbon cable I will find white connector tape and press it against the pipette as far as possible.
Now we can go to the computer and see if the pipette is connected correctly. I will click and let the robot monitor the connection. When the "green hook" appears, it means that the pipette has been successfully recognized. I want to do it without calibrating. Next exit and connect to that pipette. The initial process is the same for the base installation on the other side, next I want to connect the P20 8 channel pipette, I will start it by clicking on the three dots on the left bracket, then click Connect Pipette and Move the pipette to the front. The pipette holder will now be brought to the other side. Next, choose to connect the P20 8-channel pipette. I will attach this pipette the same way I attached the P1000 pipette to the right bracket. I also need the pipette screws that we have in the bag, the pipette doesn't have anything to hold it in right now, I need to screw in 3 pipette screws, 2 of them are embedded in the plastic case and then there's one on the bottom, so I'm going to screw on the 2 top screws first.
I need to make sure not to tighten them all the way, which I was able to do on the previous pipette, I'm going to screw in the bottom screw now because this is a multichannel pipette, so I'm not going to tighten the bottom screw all the way. First I need to go through the leveling process, but first I want to make sure that this white cable is plugged in the same way I did on the P1000, I'm going to go to the computer and make sure it's recognized, click on let the robot check the connection, it says green, instructions for pipetting The device has been recognized. I now need to adjust the pipette, I now place the calibration aluminum block in plate position 3, making sure the short side is on the left, I will lower the pipette, I can wiggle it until all pipette nozzles are touching the calibration The aluminum block, it's showing up in good shape now, so we can go back and tighten the screws, now that the pipette is connected and it's recognized and it's level, let's exit without doing the calibration.
