Pipette White Paper: Magnetic Modules

Product Introduction The Opentrons magnetic module is a module that uses magnetic bead separation technology and is used in experiments such as magnetic bead-based nucleic acid extraction. It is perfectly compatible with OT-2. The magnetic module is equipped with two independent adjustable panel supports to support standard and deep well plate laboratory equipment. The magnetic plate on the module supports a variety of different magnetic bead products, and the bracket is compatible with standard or 96-deep well plates, with volumes ranging from 10 μL to 2 mL.

Test Data In order to determine the time required to make the supernatant clear using the Opentrons magnetic module, we conducted rigorous evaluation tests. All tests are based on the default height (18mm) of 8 types of magnetic beads. Each test was performed on NEST 96-well plates. For each bead type, we will measure data in 4 volumes, namely: 20μl, 30μl, 40μl, and 50μl. Within each volume, wells A1, A6, A12, D6, E7, H1, and H12 were filled to determine how granulation changes throughout the plate. Figure 1 shows the beginning of the test (0th minute). We took images of the plate every 1 minute and observed the liquid in the wells until all wells had clear supernatants (shown in Figure 2). The results of extraction of clean beads are shown in Table 1. The results of NGS kit beads are shown in Table 2, and the library preparation results are shown in Table 3.

The deep well plate test experiments used 1ml and 2ml deep well plates respectively. Each test uses the same designated hole locations by default. In these experiments, only Omega-Biotek's mago-bind HDQ beads were used. To prepare these magnetic beads, add 400 μL HDQ Binding Buffer and 20 μL mago-bind?Particles HDQ to each well and pipet repeatedly to mix. As with the previous 96-well plate test process, we started from minute 0 (Figure 3), imaged the plate every 1 minute, and observed the wells until all wells had clear supernatants (Figure 4).

Note: When using heavier magnetic beads, such as Omega-Biotek's Mag-bound HDQ beads, be aware that these beads are heavier and will settle quickly in the test tube. Microbeads and reagents should be thoroughly mixed before use to prevent premature precipitation of magnetic beads in the supernatant, resulting in inaccurate experimental results.
