8 Lab Automation Complaints Where P.I.s Go Wrong\u2014and How to Fix Them<\/strong><\/p>\n\n\n\n

As a researcher considering lab automation, you're bound to be skeptical of the tools on the market. This is a big decision and you won't really know if it's right for you and your team until it's installed in your lab. Additionally, manufacturers make many claims: They say their tools are easy to use; they promise you won't need to learn complex programming languages \u200b\u200bor spend hours setting them up; they defend the tens (or even hundreds of thousands) of dollars you spend, Insist that the time and effort you save is worth it. This requires processing a lot of information. This book is for researchers like you who want to save time and money while improving staff productivity and the quality of your lab. We know your goals are to focus on experimental design, data analysis, writing, and publishing. We interviewed dozens of researchers who asked the same questions and expressed the same concerns you may have about lab automation\u2014and we worked hard to uncover the most common misconceptions so you can make an informed decision.<\/p>\n\n\n\n

We don\u2019t do enough pipetting to invest in automated tools. <\/strong><\/p>\n\n\n\n

Not all labs need automation\u2014automation isn\u2019t just for high-throughput labs. In 2020, liquid automation tools range in price from $5,000 to $120,000. This means that labs that perform wet lab work for less than five hours per week may not be able to justify the cost of an automated liquid handler. The best way to determine whether an automated liquid handler would be beneficial to your operation is to calculate how much time it takes to pipette in the lab. Include all pipetting steps, the logistics of moving samples in and out of the device, time spent adjusting plates and labware, and even interruptions and disturbances in the workflow. Many laboratories underestimate the amount of bench time they spend on these tasks, underestimating how often tedious tasks like preparing PCR plates, performing DNA extractions, and seeding cell culture media are completed daily, weekly, monthly, or even annually. If all this time adds up to five hours or more per week, automation can save you time, make lab staff more productive, and free up time for more intellectual lab work, like data analysis and experimental design. Some liquid automation tools are designed for low- and medium-throughput workflows. Such systems provide opportunities to laboratories that would not otherwise have the opportunity and allow laboratories the opportunity to experiment with the efficiencies of laboratory automation without budget constraints.<\/p>\n\n\n\n

Liquid handlers are not used often \u2013 it\u2019s a waste of money<\/strong><\/p>\n\n\n\n

Most laboratory equipment sits idle most of the time. But this isn't the fault of any individual liquid handler; it's often just the nature of the industry. Why might lab automation tools sit idle? As many as 25% of labs using these tools are working on highly complex projects that require time to launch, and labs may not have the time to set up these tools. Additionally, some automation solutions are only set up to perform one protocol, making it difficult or even impossible for labs to switch applications when needed. Other tools are so complex that only one or two lab members have the expertise to use them, causing all lab members to rely on these few people to keep the tools running. Frankly, some workflows simply cannot be automated, which is a hard reality to accept after robots gather dust in the lab. But in general, most labs that purchase the robot are able to successfully run the protocols they want. Some vendors even offer such a comprehensive setup process that labs can be up and running within 30 days of receiving and setting up the tool and its accessories. Compared to products and protocols of the past that were too difficult or expensive to modify, many platforms today are so flexible and open that multiple protocols can be easily customized and automated. Innovative manufacturers are providing versatile apps, support, and dedicated account managers so lab scientists don\u2019t have to be equipment technology experts\u2014they\u2019re free to do what they do best. Researchers who first determine that their workflows can truly be automated can take advantage of this useful and time-saving tool. In fact, the laboratories most successful in automating liquid handling have learned that incorporating robotics into their protocols increases efficiency, consistency, and reproducibility. They learn how to set up, calibrate, and program the tool and find the best ways to use it across multiple workflows for greater value. Because, with the right tool for the lab, research team members quickly realized that the time invested upfront in learning the tool would pay off for the entire lab.<\/p>\n\n\n\n

Manual pipetting is more accurate than liquid handlers. <\/strong><\/p>\n\n\n\n

Manual pipetting is more accurate than liquid handlers...but only if the operator uses the exact same technique every time and doesn't make any mistakes. Because even the most skilled laboratory scientists make mistakes, comparisons between manual and robotic pipetting accuracy are unrealistic. That being said, while some liquid automation tools can pipette sub-microliter volumes accurately and consistently, manual pipetting is generally recommended for volumes below one microliter (1\u00b5L). Even when used with larger volumes and properly calibrated, liquid handler pipettes may not produce consistent results - but this is often due to manual errors in calibration or protocol entry. Poor technique, use beyond the range of the pipette, inaccurate volume selection, and rapid or careless aspiration and dispensing can all lead to adverse results. For some researchers, potential inaccuracies in workflows prevent them from pursuing automation. Researchers who spend a lot of time designing workflows that require manual pipetting are often satisfied with these workflows. They know that manual pipetting can get the job done, and they prefer to avoid the discomfort and uncertainty that comes with learning how to automate. However, a good automated liquid handler can implement most of the details of manual pipetting, including speed, volume, aspiration settings, tip position, incubation time, pause steps, tip contact, and blowout\u2014every time. Once tools are properly set up and calibrated, human error is reduced and the speed, accuracy, and repeatability of experiments increase. In addition, automated liquid handling robots can further improve lab productivity and morale by reducing employees' repetitive strain injuries and reducing their exposure to hazardous materials used in certain experimental protocols.<\/p>\n\n\n\n
