Opentrons PCR \u5de5\u4f5c\u7ad9\u4e0e\u5e02\u9762\u4e3b\u6d41\u7684\u5916\u7f6e\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u76f8 \u6bd4\uff0c\u80fd\u591f\u751f\u6210\u9884\u671f\u7684PCR\u6269\u589e\u4ea7\u7269\u7684\u78b1\u57fa\u5bf9\u957f\u5ea6\u3002\u7ed3\u679c\u663e\u793a\uff0c\u65e0\u8bba\u662f\u5d4c\u5957PCR\u8fd8\u662f\u591a\u91cdPCR, \u90fd\u80fd\u4ea7\u751f\u9884\u671f\u7684 PCR\u6269\u589e\u4ea7\u7269\u7684\u78b1\u57fa \u5bf9\u957f\u5ea6\u3002<\/p>\n\n\n\n<p><strong>\u7b80\u4ecb<\/strong><br>\u7531\u4e8e\u6837\u54c1\u5236\u5907\u8fc7\u7a0b\u4e2d\u53ef\u80fd\u5b58\u5728\u6c61\u67d3\u3001\u6280\u672f\u9519\u8bef\u548c\u65f6\u95f4\u9650\u5236\uff0c\u590d\u6742\u65b9\u6848(\u4f8b\u5982\u5de2\u5f0f PCR \u548c\u591a\u91cd PCR) \u7684\u6548\u7387\u548c\u51c6\u786e\u6027\u96be\u4ee5\u4fdd\u969c\u3002\u591a\u91cd PCR \u5b9e\u9a8c\u7528\u4e8e GMO (\u8f6c\u57fa\u56e0\u751f\u7269)\u68c0\u6d4b\u3001\u6cd5\u533b\u5b66\u3001\u98df\u54c1\u5206\u6790\u3001\u7a81\u53d8\u548c\u591a\u6001\u6027\u68c0\u6d4b\u3001\u57fa\u56e0\u7f3a\u5931\u5206\u6790\u3001\u6a21\u677f\u5b9a\u91cf\u3001\u8fde\u9501\u5206\u6790\u4ee5\u53ca\u66f4\u591a\u5e94 \u7528\u3002\u8fd9\u4e9b\u591a\u91cd\u53cd\u5e94\u975e\u5e38\u9ad8\u6548\uff0c\u53ef\u5728\u5b54\u69fd\u4e2d\u4ea7\u751f\u4e24\u4e2a\u6216\u591a\u4e2a\u6269\u589e\u5b50\u7684\u4ea7\u7269\u3002<\/p>\n\n\n\n<p>\u5de2\u5f0f PCR \u662f\u53e6\u4e00\u79cd\u9ad8\u5ea6\u7279\u5f02\u6027\u7684\u6280\u672f\uff0c\u53ef\u7528\u4f5c\u6709\u7528\u7684\u8bca\u65ad\u5de5\u5177\uff0c\u7528\u4e8e\u8bc6\u522b\u751f\u7269\u6837\u54c1\u4e2d\u7684\u75c5\u539f\u4f53\uff0c\u4f8b\u5982\u52a8\u8109\u7ca5\u6837\u786c\u5316\u6591\u5757\u4e2d\u7684\u7259\u5468\u75c5 \u539f\u4f53\u3001\u8f6c\u79fb\u6027\u4e73\u817a\u764c\u7ec6\u80de\u4ee5\u53ca\u7ed3\u6838\u5206\u679d\u6746\u83cc\u548c\u9a6c\u5c14\u5c3c\u83f2\u9752\u9709\u7b49\u75c5\u6bd2\u75c5\u539f\u4f53\u4e0e\u4ec5\u9700\u8981\u4e00\u4e2a\u6269\u589e\u6b65\u9aa4\u7684\u6807\u51c6PCR \u4e0d\u540c\uff0c\u5de2\u5f0f PCR \u5177\u6709\u4e24 \u6b65\u6269\u589e\u8fc7\u7a0b\u3002\u7b2c\u4e00\u4e2a PCR \u6269\u589e\u6b65\u9aa4\u4e2d\u751f\u6210\u7684 PCR \u6269\u589e\u5b50\u7528\u4f5c\u7b2c\u4e8c\u4e2a\u6269\u589e\u6b65\u9aa4\u7684\u6a21\u677f\u3002\u8fd9\u4e9b\u590d\u5408 PCR \u5b9e\u9a8c\u53ef\u80fd\u4f1a\u56e0\u6700\u7ec8\u4ea7\u54c1\u7684\u4e0d\u540c \u800c\u53d8\u5f97\u7e41\u7410\u548c\u8017\u65f6\u3002Opentrons PCR \u5de5\u4f5c\u7ad9\u53ef\u4ee5\u81ea\u52a8\u5316\u5b9e\u73b0 PCR \u5b9e\u9a8c\uff0c\u53ea\u9700\u8981\u8fdb\u884c\u975e\u5e38\u5c11\u91cf\u7684\u624b\u52a8\u51c6\u5907\u5de5\u4f5c\uff0c\u5c31\u53ef\u4ee5\u4ea7\u51fa\u7a33\u5b9a\u7684\u4e00 \u81f4\u6027\u7ed3\u679c\u3002<\/p>\n\n\n\n<p>\u6750\u6599\u548c\u65b9\u6cd5<\/p>\n\n\n\n<p><strong>\u591a\u91cd PCR \u5b9e\u9a8c\u6b65\u9aa4<\/strong><br>\u4f7f\u7528\u4ece\u94dc\u7eff\u5047\u5355\u80de\u83cc\u548c\u91d1\u9ec4\u8272\u8461\u8404\u7403\u83cc\u5206\u79bb\u7684\u57fa\u56e0\u7ec4 DNA \u8fdb\u884c\u4e86\u591a\u91cd PCR \u5b9e\u9a8c\u3002\u6839\u636e\u5148\u524d\u7684\u7814\u7a76\uff0c\u8bbe\u8ba1\u4e86\u9488\u5bf9\u94dc\u7eff\u5047\u5355\u80de\u83cc (lasl\u3001lasR\u3001gyrB) \u548c\u91d1\u9ec4\u8272\u8461\u8404\u7403\u83cc(16s rRNA\u3001clfA\u3001mecA\u3001Eubacterial 16S rRNA)\u591a\u4e2a\u4f4d\u70b9\u533a\u57df\u7279\u5f02\u6027\u7684\u6b63\u5411\u548c\u53cd\u5411<br>\u5f15\u7269\u3002\u6bcf\u4e2a\u5b54\u9884\u671f\u4ea7\u7269\u7684\u5927\u5c0f\u5206\u522b\u4e3a600\u3001700\u548c222bp\u3002 \u5728 Opentrons OT-2 \u7684\u5e73\u53f0\u4e0a\uff0c\u57280.1 ml 96 \u5b54\u5168\u88d9\u8fb9 NEST \u677f\u4e0a\u51c6\u5907 \u4e8620 \u03bcL\u7684\u591a\u91cd PCR\u53cd\u5e94\u6df7\u5408\u7269\uff0c\u5e76\u4f7f\u7528\u8bbe\u5907\u4e0a\u7684\u70ed\u5faa\u73af\u4eea\u8fdb\u884c\u6269\u589e\u3002\u6269\u589e\u4ea7\u7269\u7ecf2%\u743c\u8102\u7cd6\u51dd\u80f6\u7535\u6cf3\u5206\u6790\u3002\u540c\u65f6\uff0c\u4e3a\u4e86\u5bf9\u6bd4\u5206\u6790\uff0c \u6211\u4eec\u4e5f\u7528\u624b\u52a8\u51c6\u5907\u4e86\u76f8\u540c\u7684\u5b9e\u9a8c\uff0c\u5e76\u5728\u7b2c\u4e09\u65b9\u624b\u52a8\u70ed\u5faa\u73af\u4eea\u4e0a\u8fd0\u884c\u3002<\/p>\n\n\n\n<p><strong>\u5de2\u5f0f PCR \u5b9e\u9a8c\u6b65\u9aa4<\/strong><br>\u5de2\u5f0f PCR \u662f\u7528\u4e8e\u9274\u5b9a\u4ece lambda \u566c\u83cc\u4f53\u5206\u79bb\u7684\u751f\u7269\u6837\u54c1\u4e2d\u7684\u75c5\u539f\u4f53\u7684\u65b9\u6cd5\u3002\u6839\u636e\u5148\u524d\u7684\u7814\u7a76\uff0c\u9488\u5bf9\u94dc\u7eff\u5047\u5355\u80de\u83cc (lasl\u3001lasR\u3001 gyrB) \u548c\u91d1\u9ec4\u8272\u8461\u8404\u7403\u83cc(16S rRNA\u3001clfA\u3001mecA\u3001Eubacterial 16S rRNA) \u8bbe\u8ba1\u4e86\u7279\u5f02\u6027\u7684\u591a\u4e2a\u4f4d\u70b9\u533a\u57df\u7684\u6b63\u5411\u548c\u53cd\u5411\u5f15\u7269\u3002 PCR\u7ec4\u5206\u88ab\u63d0\u4f9b\u7ed9OT-2 \u5e73\u53f0\uff0c\u5e76\u8f6c\u79fb\u52300.1 ml96 \u5b54 NEST\u677f\u4e2d\u3002\u5728\u6df7\u5408\u548c Opentrons \u70ed\u5faa\u73af\u6a21\u5757\u81ea\u52a8\u5bc6\u5c01\u4e4b\u524d\uff0c\u6700\u540e\u52a0\u5165 DNA \u6a21\u677f\u3002\u5bf9\u4e8e\u7b2c\u4e8c\u4e2a\u6269\u589e\u6b65\u9aa4\uff0c\u4f7f\u75280.1 ml \u7684 NEST \u5b54\u677f\u91cd\u590d\u6b64\u8fc7\u7a0b\uff0c\u5e76\u5c06\u5176\u5bc6\u5c01\u540e\u653e\u7f6e\u5728\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u4e0a\u3002\u7b2c\u4e00\u6b21\u6269\u589e\u7684\u6269\u589e\u4ea7 \u7269\u88ab\u7528\u4f5c\u9633\u6027\u5bf9\u7167\u3002<\/p>\n\n\n\n<p>PCR\u4ea7\u7269\u7ecf2%\u743c\u8102\u7cd6\u51dd\u80f6\u7535\u6cf3\u5206\u6790\u3002\u6240\u6709\u5b9e\u9a8c\u90fd\u4f7f\u7528Qubit BR\u5b9a\u91cf\uff0c\u5e76\u8ba1\u7b97\u53d8\u5f02\u7cfb\u6570\u4ee5\u8bc4\u4f30\u6269\u589e\u7684\u4e00\u81f4\u6027\u3002<\/p>\n\n\n\n<p><strong>\u5b9e\u9a8c\u7ed3\u679c<\/strong><br>\u4f7f\u7528Opentrons \u70ed\u5faa\u73af\u6a21\u5757\u5728OT-2\u4e0a\u8fdb\u884c\u81ea\u52a8\u5316\u591a\u91cdPCR\u5b9e\u9a8c\uff0c\u8fd0\u884c\u7ed3\u679c\u663e\u4e0e\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u76f8\u5f53\u3002<\/p>\n\n\n\n<p>\u591a\u91cd PCR\u7684\u5b9e\u9a8c\u5e03\u5c40\u91c7\u7528\u4e86 Kim \u7b49\u4eba\u57282018\u5e74\u7684\u7814\u7a76\u4e2d\u7684\u65b9\u6cd5\u3002\u6837\u54c1\u5b54\u4f4d\u6807\u8bb0\u4e3a\u7eff\u8272\uff0c\u9634\u6027\u5bf9\u7167\u6807\u8bb0\u4e3a\u84dd\u8272(\u89c1\u56fe1)\u3002<\/p>\n\n\n\n<figure \u4ea7\u7269\u68c0\u6d4b\u3002\u4e0e\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u751f\u6210\u7684PCR \u4ea7\u7269\u76f8\u6bd4(\u56fe2B), OT-2\u70ed\u5faa\u73af\u4eea\u4e0a\u751f\u6210\u7684 PCR \u6269\u589e\u4ea7\u7269\u4e0d\u4ec5\u663e\u793a\u51fa\u66f4\u6e05\u6670\u548c\u660e\u663e\u7684\u6761\u5e26\uff0c\u800c\u4e14\u957f\u5ea6\u7b26\u5408\u9884\u671f (lasl\u4e3a600bp,lasR \u4e3a700bp,gyrB \u4e3a222bp) ( \u56fe 2A)\u3002<br>\u4e3a\u4e86\u8fdb\u4e00\u6b65\u8bc1\u660e OT-2 \u7684\u6548\u7387\uff0c\u4f7f\u7528\u9488\u5bf9\u91d1\u9ec4\u8272\u8461\u8404\u7403\u83cc(16s rRNA \u3001clfA \u3001mecA \u3001Eubacterial 16s rRNA) \u7684\u7279\u5f02\u6027\u5f15\u7269\u8fdb\u884c \u4e86\u81ea\u52a8\u5316\u591a\u91cd PCR\u3002\u9884\u671f\u7684\u78b1\u57fa\u5bf9\u957f\u5ea6\u5206\u522b\u4e3a791bp\u3001638bp\u3001499bp \u548c371bp, \u5e76\u5728\u743c\u8102\u7cd6\u51dd\u80f6\u4e0a\u68c0\u6d4b(\u56fe2C) \u3002 \u540c\u65f6\uff0c\u4f7f \u7528\u76f8\u540c\u7684PCR\u7ec4\u5206\u8fdb\u884c\u4e86\u624b\u52a8\u591a\u91cd PCR\u534f\u8bae\uff0c\u4f7f\u7528\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u8fdb\u884c\u64cd\u4f5c(\u56fe2)\u3002\u5728 OT-2\u4e0a\u81ea\u52a8\u5316\u548c\u624b\u52a8\u8fdb\u884c\u7684\u591a\u91cd PCR\u90fd \u663e\u793a\u51fa\u4e86\u7c7b\u4f3c\u7684\u6269\u589e\u6548\u679c(\u56fe2A\u30012B)\u3002<\/p>\n\n\n\n<figure class=\"wp-block-image size-full is-resized\"><img decoding=\"async\" \u9053\u663e\u793a\u4e86\u5728\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u4e0a\u624b\u52a8\u52a0\u6837\u540e\u7684\u4ea7\u7269\u3002<\/p>\n\n\n\n<p>\u4e3a\u4e86\u9a8c\u8bc1\u5728OT-2 \u548c\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u4e0a\u751f\u6210\u7684 PCR \u7684\u7cbe\u786e\u6027\uff0c\u5206\u6790\u4e86\u53d8\u5f02\u7cfb\u6570 (CV%), \u7ed3\u679c\u663e\u793a\u5728\u591a\u91cd PCR \u548c \u5de2 \u5f0f PCR \u5b9e\u9a8c\u4e2d\uff0c OT-2 \u7684 CV% \u503c\u4e0e\u7b2c\u4e09\u65b9\u70ed\u5faa\u73af\u4eea\u76f8\u6bd4\u76f8\u540c\u6216\u8f83\u4f4e\uff0c\u8868\u660e\u6269\u589e\u7684\u5747\u5300\u6027\u66f4\u597d(\u89c1\u88681)\u3002<\/p>\n\n\n\n<p>\u88681:\u5728\u4e0d\u540c\u6a21\u677f\u7c7b\u578b\u548cPCR\u5b9e\u9a8c\u4e2d\u8868\u73b0\u51fa\u9ad8\u5ea6\u5747\u5300\u7684\u6269\u589e\u6548\u679c\u3002<br>CV\u503c(\u53d8\u5f02\u7cfb\u6570)\u4e3a\u7ecf\u8fc7Qubit \u5b9a\u91cf\u540e\u8ba1\u7b97\u5f97\u51fa\u7684\u7ed3\u679c\uff0c\u5206\u522b\u663e\u793a\u4e86\u5728OT-2
